Human CYP1A2 metabolizes organophosphorus compounds, and is also inhibited by them
- LC-MS/MS metabolism assays and a fluorescence inhibition assay were used to test how human CYP1A2 handles organophosphorus pesticides and nerve agent surrogates.
- Thion forms were turned over more readily than oxon forms, and several compounds inhibited the enzyme in a time- and concentration-dependent manner.
- Docking on a CYP1A2 crystal structure was consistent with the experimental trends.
The time-dependent inhibition is what catches my eye: an exposure that quietly inactivates CYP1A2 would shift the clearance of co-administered substrates such as caffeine, theophylline or clozapine. The bioscavenger idea is a stretch from recombinant enzyme work, and I'd want hepatocyte data and realistic exposure levels before reading anything into clinical or occupational risk.
