Incyte's G12D Inhibitor Posts 89% Response With FOLFIRINOX in Pancreatic Cancer
- In a Phase 1 study of 54 metastatic pancreatic cancer patients, Incyte's KRAS G12D inhibitor plus mFOLFIRINOX produced an 89% confirmed response rate; with gemcitabine/nab-paclitaxel it was 69%.
- Safety was described as manageable and in line with chemotherapy alone.
- Analysts see it as competitive with Revolution Medicines' zoldonrasib (82% and 61% in comparable regimens); a first-line Phase 3 is underway.
A 7-point gap over zoldonrasib's 82% in the FOLFIRINOX setting is noise in a cross-trial comparison of a 54-patient Phase 1. The real signal is that Incyte is now a credible second contender in G12D pancreatic cancer, not a leader. With a first-line Phase 3 already running, progression-free survival and tolerability will decide the race, and this readout doesn't answer either.
