M-FRAG embeds molecules and fragments together so property-relevant fragments can guide reinforcement-learning molecule assembly
- M-FRAG aligns molecules and fragments in one embedding space, organizing fragments by their relationship to parent molecules and by how they distribute across property values.
- Fragments near property-relevant regions are prioritized and used to steer reinforcement-learning-based molecular assembly.
- Evaluated computationally on five docking-based multiobjective optimization tasks, it yielded more novel hits and better top docking scores than baselines while keeping novelty and validity; no experimental testing.
Docking scores are the only reward here, and generators are notorious for exploiting scoring-function quirks, so better top scores say little about real binders. The useful question is whether property-aware fragment selection beats a plain fragment-frequency prior at equal sampling budget. Rescoring the hits with a different method or a free-energy calculation, plus a synthesizability check, would show whether they are real.
