Peroxynitrite opens TRPV1 to drive tetrathiomolybdate-induced muscle growth
- Transcriptome analysis and functional tests identified TRPV1 as a required mediator of the muscle hypertrophy triggered by tetrathiomolybdate-generated superoxide.
- TRPV1 knockout abolished the effect and TRPV1 activation mimicked it; superoxide raised NOS activity and nitric oxide, which formed peroxynitrite that directly activates the channel.
- Peroxynitrite scavengers blocked hypertrophy even with superoxide and nitric oxide unchanged, placing peroxynitrite as the specific trigger.
💡 Points Based on the abstract
The knockout-plus-scavenger logic is tidy, but muscle growth by an oxidant should make a pharmacologist nervous: TRPV1 activation also drives pain, heat sensitivity and calcium overload in damaged fibres. Whether the same axis is protective in disease muscle or just a physiological adaptation is untested. A dose-response of exposure against hypertrophy versus toxicity would show if there's any therapeutic window.
