Target-ID work points to calpain-2 as how sodium danshensu protects the heart after reperfusion
- Sodium danshensu, a salvianic-acid derivative, was traced to calpain-2 as its direct binding partner using protein-array and mass-spec proteomics, thermal-shift and protease-stability assays, plus docking.
- Calpain-2 is abundant in heart tissue, and pathway analysis tied its activity to cardiomyocyte apoptosis after reperfusion.
- In mouse artery-ligation and cell hypoxia-reoxygenation models, the compound lessened injury, and a selective calpain-2 inhibitor produced a comparable protective effect.
💡 Points Based on the abstract
Does a selective calpain-2 inhibitor mimicking the compound prove the mechanism? Not alone. Calpain-1 and -2 are notoriously hard to separate pharmacologically, and the missing experiment is danshensu in CAPN2-knockdown or knockout hearts: if protection vanishes, the target is real. I'd also want binding affinity and cardiac exposure at efficacious doses before calling it a lead.
