Chk1 inhibition eases motor deficits in mouse spinal cord injury
- Chk1 was tested as a driver of microglial M1 polarization and pyroptosis after spinal cord injury.
- Mouse contusion injury and LPS-treated BV2 microglia were used; Chk1 down-regulation reversed the polarization shift and pyroptosis and improved motor function, while up-regulation worsened both.
- In cells, LPS raised DNA damage (γ-H2AX) and Chk1 activation; the inhibitor SCH900776 and repair agonist AV-153 were used to probe the link. The abstract is truncated.
💡 Points
Genetic knockdown and overexpression moving outcomes in opposite directions is good target-validation logic, but it isn't a drug result. SCH900776 was built for oncology, so CNS exposure after contusion is the first thing I'd check. BV2 cells are a weak stand-in for in vivo microglia, and Chk1 blockade risks collateral DNA-damage toxicity.
