New 5-arylpyrazole chemotype blocks MCT1 lactate transport

  • A small set of new 5-arylpyrazoles was made via a multistep route, offering a fresh scaffold against MCT1, a lactate transporter tied to tumor metabolism.
  • In a 3-bromopyruvate-based transport screen, the 3-fluorophenyl analogue 17c curbed MCT1 activity at concentrations below those that harmed cells.
  • 17c also caused intracellular lactate to build up across several cell lines, consistent with transporter blockade, and is proposed as a lead for further optimization.
💡 Points Based on the abstract

Eleven compounds and one hit with lactate accumulation below cytotoxic doses is a reasonable start, but no binding data means I can't say it's on-target MCT1 rather than a general transport or metabolic effect. Low-yield multistep chemistry would also slow SAR. I'd first test MCT4 selectivity and compare against AZD3965-type chemotypes.
