TRPV3 turns out to be a driver of lung inflammation in mice
- TRPV3 expression rose in the lungs of mice with LPS- or carvacrol-induced acute lung injury, and activating the channel with carvacrol alone reproduced features of the injury.
- Genetic deletion or pharmacological inhibition of TRPV3 attenuated the lung injury.
- In alveolar epithelial cells, TRPV3 activation raised intracellular calcium, boosted cytokine and chemokine output and loosened tight junctions; TNF-α enhanced TRPV3 calcium responses, suggesting a feed-forward loop.
💡 Points Based on the abstract
Knockout, inhibitor and agonist all pointing the same way is a decent validation package for an ion channel. The gap is that carvacrol isn't selective for TRPV3 (it hits TRPA1 too), and the inhibitors in this field have a patchy selectivity record, so the genetic arm carries the weight. TRPV3 antagonists are being pursued for skin, which makes lung safety and exposure an obvious next question.
