CCR8xCD3 T cell engager clears tumor-infiltrating Tregs and slows growth in mouse tumor models
- A bispecific antibody links CD3 on T cells to CCR8, which is enriched on tumor-infiltrating Tregs, so T cells rather than NK cells carry out the depletion.
- It selectively removed Tregs in human solid tumor samples in vitro.
- In several syngeneic mouse models with differing T cell infiltration, it slowed tumor growth, alone or alongside anti-PD-L1.
💡 Points Based on the abstract
Swapping NK-mediated ADCC for T cell redirection is the bet here, and it's a double-edged one: CD3 engagers bring cytokine release and on-target hits on any CCR8+ activated T cell. Syngeneic wins say little about human Treg biology, since mouse CCR8 expression differs. Without the primate PK and tolerability data, I wouldn't move forward.
