Retooling imatinib's tail to make a Lyn-selective kinase probe

  • Lyn is hard to target selectively because its Src-family relatives look so alike, so the authors varied imatinib's tail region to favor it.
  • Top analogs are potent on Lyn, with one strongly selective over Hck and another staying clean even at high ATP.
  • In human microglia the lead blunted amyloid-triggered Lyn autophosphorylation and shifted microglial states. Preprint, not yet peer reviewed.
💡 Points Based on the abstract

Tail-only changes on a known type II scaffold are a sound call: cheap to make, and they exploit DFG-out and activation-loop differences rather than fighting the near-identical hinge pocket. The Hck selectivity figure deserves a stress test. Worth checking: a full Src-family panel, a kinome scan, and whether the 3D-QSAR predicts held-out analogs.
