Small molecule gramine shields liver cells from alcohol damage by driving down NOX1

  • Gramine was nominated as a NOX1-binding compound; docking and a thermal shift assay pointed to direct engagement, and it hastened NOX1 protein turnover.
  • In ethanol-stressed hepatocytes, knocking down NOX1 or RAC1 reproduced much of the benefit, easing oxidative, mitochondrial and ferroptosis-type damage.
  • Gramine improved cell health even when given after ethanol, but forcing NOX1 expression weakened its effect.

Knockdown phenocopying the drug is reassuring, but it is cell-only; I'd want Nox1-null or hepatocyte-specific mice under chronic-plus-binge ethanol before going further. Also, docking plus a thermal shift on a ~160 Da indole is thin proof of binding, and with no free-exposure or selectivity data, a plant alkaloid with known toxicity could easily be working off-target.
