VCAM-1-targeted nanoparticles deliver a CB2 agonist to plaques, with sex-dependent benefit
- The CB2 agonist JWH-133 was loaded into PLGA nanoparticles decorated with a VCAM-1-binding peptide to concentrate it at inflamed plaque endothelium.
- In male and female ApoE-deficient mice the targeted particles reduced foam-cell-rich deposits and shifted inflammatory markers, outperforming free drug.
- Benefit was larger in males, who had greater baseline plaque burden.
💡 Points Based on the abstract
Sex-specific outcomes are reported, but with a baseline plaque imbalance the 'bigger effect in males' may just reflect more disease to reduce. What's missing is biodistribution and PK of particle versus free drug, and evidence the benefit is CB2-mediated (a CB2 antagonist or knockout arm), plus a comparison against a non-targeted particle. JWH-133 is also a tool compound, so a clinical-grade agonist would be the real test.
