Corydamine analogs hit glutathione reductase to boost sorafenib in liver cancer
- A series of corydamine-based analogs was built, and lead 8g matched sorafenib in HCC cell activity.
- 8g appears to engage glutathione reductase, upsetting glutathione turnover and NADPH balance and undermining tumor redox defenses.
- Paired with sorafenib, it raised ROS, curbed migration and invasion, and triggered G2/M arrest and apoptosis, giving synergy.
- The compound showed good microsomal stability, low hERG liability and high oral exposure, and the combination outperformed sorafenib alone in a Huh7 xenograft.

"Appears to engage" is carrying a lot here. Before I believed glutathione reductase is the driver, I'd want a cellular target-engagement readout, a GR-knockdown phenotype, and counterscreens, since a natural-product-derived scaffold with a redox-active motif can generate ROS nonspecifically. Also check whether 8g is simply a covalent or redox-cycling nuisance. Then see if the synergy holds in sorafenib-resistant lines.
