Hastatoside protects mouse liver from alcohol injury through the ATF4–ZBP1 PANoptosis axis

  • In mice on a 28-day ethanol diet, hastatoside lowered ATF4 and PANoptosis-related sensors in liver and reduced injury.
  • ATF4 silencing cut ZBP1 and protected the liver, and a reporter assay linked ATF4 to the ZBP1 promoter; hepatocyte-specific ATF4 loss made the compound's effect stronger.
  • In macrophage–hepatocyte co-culture, hastatoside also suppressed macrophage pyroptosis and the inflammation it feeds.
💡 Points Based on the abstract

Note the genetics: if hepatocyte ATF4 deficiency already gives the benefit, the compound adds on top of it, which is awkward for a claim that ATF4 is its mediator. Plant glycosides are also often poorly absorbed, so unlike a gene knockdown the question is what reaches the liver. Oral PK and a comparison with an accepted reference such as an anti-inflammatory benchmark would help me trust the efficacy size.
